== Adverse effects == People have had severe infusion-related reactions to pembrolizumab. There have also been severe immune-related adverse effects including lung inflammation (including fatal cases) and inflammation of endocrine organs that caused inflammation of the pituitary gland, of the thyroid (causing both hypothyroidism and hyperthyroidism in different people), and pancreatitis that caused Type 1 diabetes and diabetic ketoacidosis; some people have had to go on lifelong hormone therapy as a result (e.g. insulin therapy or thyroid hormones). People have also had colon inflammation, liver inflammation, kidney inflammation due to the drug. The common adverse reactions have been fatigue (24%), rash (19%), itchiness (pruritus) (17%), diarrhea (12%), nausea (11%) and joint pain (arthralgia) (10%). Other adverse effects occurring in between 1% and 10% of people taking pembrolizumab have included anemia, decreased appetite, headache, dizziness, distortion of the sense of taste, dry eye, high blood pressure, abdominal pain, constipation, dry mouth, severe skin reactions, vitiligo, various kinds of acne, dry skin, eczema, muscle pain, pain in a limb, arthritis, weakness, edema, fever, chills, myasthenia gravis, and flu-like symptoms.
=== Electricity demand and grid capacity === The proliferation of AI has driven a surge in data centre construction and power requirements. === Urogynaecological === Weakened connective tissues can lead to pelvic organ prolapse in female patients with EDS. === Macrophages === One of the roles of macrophages is to phagocytize other expended phagocytes, bacteria and damaged tissue, and they also debride damaged tissue by releasing proteases. Macrophages function in regeneration and are essential for wound healing. They are stimulated by the low oxygen content of their surroundings to produce factors that induce and speed angiogenesis and they also stimulate cells that reepithelialize the wound, create granulation tissue, and lay down a new extracellular matrix. By secreting these factors, macrophages contribute to pushing the wound healing process into the next phase. They replace PMNs as the predominant cells in the wound by two days after injury. The spleen contains half the body's monocytes in reserve ready to be deployed to injured tissue. Attracted to the wound site by growth factors released by platelets and other cells, monocytes from the bloodstream enter the area through blood vessel walls. Numbers of monocytes in the wound peak one to one and a half days after the injury occurs. Once they are in the wound site, monocytes mature into macrophages. Macrophages also secrete a number of factors such as growth factors and other cytokines, especially during the third and fourth post-wounding days. These factors attract cells involved in the proliferation stage of healing to the area. In wound healing that result in incomplete repair, scar contraction occurs, bringing varying gradations of structural imperfections, deformities and problems with flexibility. === Telomere shortening === Telomeres are nucleoid sequences that repeat themselves and cap chromosomes. They protect chromosomes from dilapidation and recombination abnormalities. Their length decreases with every division of the cell and results in cellular senescence. They are critical structures at the end of the eukaryotic chromosomes, consisting of many copies of G rich repeats. Without telomeres, chromosomes will combine and cause instability in the genes. The enzyme that increases telomere length to prevent them from becoming short is called telomerase. Deficiency of this enzyme can hasten telomere shortening which can cause a flawed regeneration of the tissue. This also suppresses the production of epidermal cells. Also, exposure to UV radiation causes mutations to telomeres and sufficient exposure can result in the deaths of cells.
