This is a working overview of MC1R, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-06-12 and is reviewed periodically as new material appears.
Receptor binding at MC1R on melanocytes raises intracellular cyclic AMP and increases expression of tyrosinase and related enzymes. The downstream result is greater synthesis of eumelanin, the dark pigment, without ultraviolet exposure acting as the trigger. The compound is not selective, however, and also engages MC3R, MC4R and MC5R, which are expressed in the central nervous system and elsewhere. That lack of selectivity is the explanation usually offered for effects reported outside pigmentation, including appetite suppression and nausea. Selectivity remains a central theme in comparative studies of related peptides.
Human data remain limited and mostly short-term. Reports describe small trials and observational accounts rather than large controlled studies, so questions about dose-response relationships and long-term effects on melanocytes stay open. Whether repeated exposure alters naevus behaviour is not settled in the published record. Researchers also note that self-administered use outside clinical settings makes actual exposure difficult to quantify. Statements about efficacy and safety should therefore be read as preliminary rather than established.
Melanotan II is a synthetic cyclic heptapeptide that acts as an agonist at melanocortin receptors. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, the endogenous peptide involved in pigment production. The analogue carries a lactam bridge that constrains the ring and slows enzymatic breakdown relative to the native hormone. In research literature it appears under several abbreviations, and naming conventions are not fully standardized. Published descriptions usually place it within the broader melanocortin agonist family.
Melanotan II is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its structure substitutes a lactam bridge between side chains to increase stability relative to the native hormone. The compound is also known by the shorthand MT-II and by several non-proprietary synonyms used in research catalogues. It is not an approved therapeutic product in any major jurisdiction; material sold under this name is typically offered as a laboratory reagent rather than as a medicine.
Activity is attributed to agonism at melanocortin receptors, particularly MC1R and MC4R. Activation of MC1R on melanocytes increases melanin synthesis, which underlies the reported tanning effect. MC4R engagement in the central nervous system is linked to appetite suppression and to effects on sexual arousal reported in early clinical studies. Those studies were small and were not designed to establish efficacy or long-term safety. Receptor selectivity among the melanocortin subtypes is not absolute, which complicates attribution of any effect to a single pathway.

An immobilized enzyme is an enzyme, with restricted mobility, attached to an inert, insoluble material—such as calcium alginate (produced by reacting a mixture of sodium alginate solution and enzyme solution with calcium chloride). This can provide increased resistance to changes in conditions such as pH or temperature. It also lets enzymes be held in place throughout the reaction, following which they are easily separated from the products and may be used again - a far more efficient process and so is widely used in industry for enzyme catalysed reactions. An alternative to enzyme immobilization is whole cell immobilization. Immobilized enzymes are easily to be handled, simply separated from their products, and can be reused.
== Partition coefficient and log P == The partition coefficient, abbreviated P, is defined as a particular ratio of the concentrations of a solute between the two solvents (a biphase of liquid phases), specifically for un-ionized solutes, and the logarithm of the ratio is thus log P. When one of the solvents is water and the other is a non-polar solvent, then the log P value is a measure of lipophilicity or hydrophobicity. The defined precedent is for the lipophilic and hydrophilic phase types to always be in the numerator and denominator respectively; for example, in a biphasic system of n-octanol (hereafter simply "octanol") and water:
== Substrate recognition == Ubiquitin ligases are the final, and potentially the most important determinant of substrate specificity in ubiquitination of proteins. The ligases must simultaneously distinguish their protein substrate from thousands of other proteins in the cell, and from other (ubiquitination-inactive) forms of the same protein. This can be achieved by different mechanisms, most of which involve recognition of degrons: specific short amino acid sequences or chemical motifs on the substrate.

Since irregularities in the menstrual cycle are normal shortly after menarche and during perimenopause, the age of the individual and the time since menarche need to be taken into account. Menstrual cycles are considered irregular in the following cases:
Abraham and Chain discovered that some airborne bacteria produced penicillinase, an enzyme that destroys penicillin. It was not known why the mould produced penicillin, as the bacteria penicillin kills are no threat to the mould; it was conjectured that it was a byproduct of metabolic processes for other purposes. The next stage of the process was to extract the penicillin. The liquid was filtered through parachute silk to remove the mycelium, spores and other solid debris. The solution was acidified by the addition of phosphoric acid for the dissociation process. Chain determined that penicillin was stable only with a pH of between 5 and 8, but the process required one lower than that. By keeping the mixture at 0 °C, he could retard the breakdown process. In this form the penicillin could be drawn off by a solvent. Initially ether was used, as it was the only solvent known to dissolve penicillin, but it is highly inflammable and toxic. At Chain's suggestion, they tried using the much less flammable amyl acetate instead, and found that it also worked.
In 1973, Cold Storage's Pasaraya Malaysia, a joint venture of Cold Storage's subsidiary Supermarkets Malaysia and "Malaysian interests" was responsible for the construction of Jaya Supermarket in Petaling Jaya, set to be completed in April 1974. In 1985, Cold Storage Malaysia opened its first Majik Market store in Bangsar Baru, Kuala Lumpur, It operated under a franchise from Mumford Inc of Atlanta, USA, which complements its existing Jaya Supermarket chain but differ in concept and management. Due to competition from 7-Eleven, whose Malaysian franchise was then operated by the Antah Holdings group, Cold Storage Malaysia phased out Majik Market as part of its streamlining process. Cold Storage Malaysia, along with Peremba, Gaya Timur and Jusco were responsible for the initial version of Jaya Jusco "superstores" in 1985, with three stores in Dayabumi, Taman Tun Dr Ismail and Subang Jaya. In 1987, Cold Storage Malaysia, which had been through a difficult financial phase in the previous three years, had a change in management with some senior executives leaving. In 1992, DFI Retail Group purchased the company, although no major changes were made to the Cold Storage brand. Cold Storage started the Fresh Food Distribution Centre in Singapore in 1999, a composite multi-temperature warehousing for fresh and frozen food distribution. In 2023, DFI Retail Group divested its Malaysian food retailing business (including Cold Storage brand in that country) to Macrovalue Sdn Bhd.

It is a synthetic cyclic heptapeptide and a non-selective melanocortin receptor agonist. Structurally it is modelled on alpha-melanocyte-stimulating hormone, a peptide that occurs naturally in the body.
Activation of MC1R increases pigment synthesis independently of ultraviolet exposure, which is the mechanism usually cited for darkening without sun exposure. Ultraviolet light still influences skin response through separate biological pathways.
Because the peptide binds several melanocortin receptor subtypes, its effects are not confined to pigmentation. This complicates interpretation of both intended and unintended outcomes reported in studies.
No. No major regulatory agency has granted a marketing authorisation for melanotan II as a medicine. Products sold under this name are generally presented as laboratory reagents and are not subject to the batch-release testing applied to approved drugs.
