Melanocortin receptors comprise five subtypes with distinct tissue distributions and functions. Melanotan-2 is described in the literature as a non-selective agonist that engages several of these subtypes, including MC1R, MC3R, MC4R, and MC5R. MC1R is the subtype most directly linked to melanin production in skin cells. Because the compound is not subtype-selective, its observed effects in experimental settings are generally attributed to activity across multiple receptor pathways rather than to a single target.
Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
The compound emerged from research programs in the 1980s that examined analogues of alpha-melanocyte-stimulating hormone for pigmentation and photoprotection. Investigators modified the native sequence to extend activity duration and potency. A related analogue, afamelanotide, was developed within the same broad line of inquiry and eventually gained approval in certain jurisdictions for a rare light-sensitivity condition. Melanotan-2 itself did not progress through the same regulatory route and has no approved therapeutic indication.
Solid peptide kept dry at minus twenty degrees Celsius, shielded from light and moisture, is generally considered stable for extended periods. Solutions are divided into single-use aliquots and held at minus twenty or minus eighty degrees Celsius, because repeated freeze-thaw cycles promote aggregation and loss of material to container surfaces. Hydrolysis of the backbone and oxidation of tryptophan are the principal degradation routes in aqueous solution, and both accelerate at ambient temperature. Hygroscopic uptake after a vial is opened can also shift the actual mass weighed, which affects any concentration calculated from it.
Routine characterisation relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nanometres, using a C18 column and a water-acetonitrile gradient containing trifluoroacetic acid. Electrospray ionisation mass spectrometry confirms the expected molecular mass and can reveal truncated or oxidised by-products that co-elute poorly. Sequence and stereochemistry require additional work, such as peptide mapping or amino acid analysis, because a chromatographic purity figure alone does not distinguish a diastereomer from the target peptide. Independent testing of research-grade material frequently shows measured content below the stated label, so a certificate of analysis is best read together with the method that produced it.
Melanotan-2 is handled in the laboratory as a lyophilised powder that dissolves readily in water, dimethyl sulfoxide and dimethylformamide, with limited solubility in ethanol. Stock solutions prepared in an organic solvent often precipitate when diluted into aqueous buffer, so gradual dilution with mixing is standard practice. The peptide carries a tryptophan residue and a histidine residue, both sensitive to oxidation and to alkaline conditions. Working solutions are therefore kept near neutral to slightly acidic pH, protected from light, and consumed within the same working session whenever that is practical.

Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
Lyophilised peptide powder is comparatively stable when kept dry, cold and protected from light. Once dissolved, the molecule is exposed to hydrolysis, oxidation and microbial growth, and degradation accelerates at higher temperatures and in alkaline solution. Repeated freeze-thaw cycles concentrate solutes and promote aggregation. Handling guidance for research peptides commonly clusters around freezer temperatures for powder and short refrigerated use for reconstituted solutions, with pH control and sterile technique applied throughout.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
Analytical confirmation of identity relies on mass spectrometry, most often coupled to liquid chromatography. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and provides a purity estimate based on peak area. Electrospray ionization mass spectrometry then confirms the expected molecular mass, while tandem mass spectrometry can map the fragment sequence. For research-grade material, these two techniques together form the standard minimum. Purity figures reported by vendors are frequently not traceable to an independent laboratory.
Independent verification is central to quality control because the compound is not produced under pharmaceutical manufacturing standards. Third-party laboratories can measure purity, identity, residual solvents, and microbial contamination, though the scope of testing varies between services. Reported analyses of vendor samples have shown batch-to-batch variation in peptide content and the presence of truncated or oxidized species. How much of this variation reflects synthesis conditions versus storage and shipping is not well characterized. No harmonized reference standard exists for the material as sold.
The Claisen-Schmidt condensation between 3-(methoxymethoxy)benzaldehyde [13709-05-2] (1) and cyclohexanone (2) afforded CID:54364197 (3). Michael addition of dimethylamine leads the aminoketone, i.e. 2-[dimethylamino-[3-(methoxymethoxy)phenyl]methyl]cyclohexan-1-one, CID21518320. Reduction of the ketone proceeds stereospecifically to afford the cis aminoalcohol [51356-58-2] (4). Mild hydrolysis of the product gives the free phenol ciramadol (5).
=== Discontinued menu items === One of Taco Bell's original 1962 menu items was the Chiliburger, renamed the Bell Burger in 1969, then the Bell Beefer in 1979; this was a loose meat sandwich originally filled with taco-seasoned ground beef in mild red sauce with shredded lettuce and diced onion served on a steamed hamburger bun. Later, shredded cheese and diced tomato were added to the sandwich. Taco Bell discontinued the Bell Beefer around 1986 to maintain a more typical Tex-Mex-inspired menu. Other discontinued Taco Bell menu items include the Enchirito (name revived for a different menu item); Taco Lite; Taco Grande; Chilito (Chili Cheese Burrito); Beefy Crunch Burrito; Beefy Melt Burrito; Seafood Salad; Chicken Fiesta Burrito; Potatorito; Volcano Taco; BLT Taco; Cheesarito; Cinnamon Crispas; Nacho Crunch Grilled Stuft Burrito; Chicken Caesar Grilled Stuft Burrito; Grilled Stuft Nacho; Fully Loaded Nachos; Crunchwrap Sliders; Blackjack Taco; Bean Burrito Especial; Border Ices; and the Meximelt. In September 2019, Taco Bell made new changes to its menu. Items discontinued include the Beefy Mini Quesadilla; Chips and Salsa; Chipotle Chicken Loaded Griller; Double Decker Taco; Cool Ranch and Fiery Doritos Locos Tacos; Double Tostada; Power Menu Burrito, and the XXL Grilled Stuft Burrito.
== Description == One of the beautiful, intelligent and smart fish,"The King betta" generally grows to about 5–6 cm (2–2 1/2 in). Its pigments are generally vibrant beige to a white. As with most bettas, King betta females are smaller (even the fins) and less colorful than the males. The king betta can be happy or even aggressive based on the environment and can be trained easily (rather than the wild bettas') and they remember their owners for longer time period. The life span of the King betta is short, generally 2–3 years, rarely reaching four. The King betta is (as are all other bettas) members of the gourami family (Osphronemidae) and prefers a temperature of 78–82 °F.
Cyanobacteria are the simplest organisms that have been observed demonstrating circadian rhythms.(2)(3) The primitiveness and simplicity make the KaiC phosphorylation model invaluable to circadian rhythm research. While it is much simpler than models for eukaryotic circadian rhythm generators, the principles are largely the same. In both systems the circadian period is dependent on the interactions between proteins within the cell, and when the genes for those proteins are mutated, the expressed period changes. (1)(2) This model of circadian rhythm generation also has implications for the study of circadian “evolutionary biology”. Given the simplicity of cyanobacteria and of this circadian system, it may be safe to assume that eukaryotic circadian oscillators are derived from a system similar to that present in cyanobacterium. where M is molar mass of the radionuclide, and NA is the Avogadro constant. Practically, the mass number A of the radionuclide is within a fraction of 1 % of the molar mass expressed in g/mol and can be used as an approximation. Specific radioactivity a is defined as radioactivity per unit mass of the radionuclide:
By binding tightly to partially synthesized peptide sequences (incomplete proteins), Hsp70 prevents them from aggregating and being rendered nonfunctional. Once the entire protein is synthesized, a nucleotide exchange factor (prokaryotic GrpE, eukaryotic BAG1 and HspBP1 are among those which have been identified) stimulates the release of ADP and binding of fresh ATP, opening the binding pocket. The protein is then free to fold on its own, or to be transferred to other chaperones for further processing. HOP (the Hsp70/Hsp90 Organizing Protein) can bind to both Hsp70 and Hsp90 at the same time, and mediates the transfer of peptides from Hsp70 to Hsp90. Hsp70 also aids in transmembrane transport of proteins, by stabilizing them in a partially folded state. It is also known to be phosphorylated which regulates several of its functions. Hsp70 proteins can act to protect cells from thermal or oxidative stress. These stresses normally act to damage proteins, causing partial unfolding and possible aggregation. By temporarily binding to hydrophobic residues exposed by stress, Hsp70 prevents these partially denatured proteins from aggregating, and inhibits them from refolding. Low ATP is characteristic of heat shock and sustained binding is seen as aggregation suppression, while recovery from heat shock involves substrate binding and nucleotide cycling. In a thermophile anaerobe (Thermotoga maritima) the Hsp70 demonstrates redox sensitive binding to model peptides, suggesting a second mode of binding regulation based on oxidative stress.
=== The Pékerman era and the golden generation (2011–2018) === At the 2011 Copa América in Argentina, Colombia won Group A ahead of the hosts with seven points and without conceding, beating Costa Rica 1–0 and Bolivia 2–0 either side of a goalless draw with Argentina, but were eliminated 2–0 by Peru in the quarter-finals at Córdoba after extra time. José Pékerman, the Argentine who had taken his own country to the 2006 World Cup quarter-finals, was appointed head coach in January 2012. Colombia finished second in the CONMEBOL qualifiers with 30 points to reach the 2014 FIFA World Cup, ending a sixteen-year absence, and conceded only 13 goals — the second-best defensive record in the group behind Argentina.
== Brewing == While colloquially named “tea,” peppermint tea is actually an infusion, or tisane. True teas (such as black, green, oolong, and white tea) are all derived from the Camellia sinensis plant, while tisanes are made from the flowers, leaves, roots, seeds, bark, or berries of a variety of different plants. Like true teas, mint tea may be consumed hot or cold, and served with milk or sweeteners such as sugar or honey. True teas contain caffeine and tannins, compounds that can produce a bitter taste. The levels of these compounds in a cup of tea can vary based on steeping time and water temperature. To achieve a pleasant drinking experience, true teas are prepared using specific brewing times and temperature ranges. Mint tea, as a tisane, does not contain these compounds at similarly high concentrations, making it less prone to bitterness from oversteeping, allowing for a wider range of steeping times and temperatures.
=== Filaments === Nylon filaments are primarily used in brushes especially toothbrushes and string trimmers. They are also used as monofilaments in fishing line. Nylon 610 and 612 are the most used polymers for filaments. Its various properties also make it very useful as a material in additive manufacturing; specifically, as a filament in consumer and professional grade fused deposition modeling 3D printers.

== Clinical relevance == Pyruvate dehydrogenase deficiency (PDCD) can result from mutations in any of the enzymes or cofactors used to build the complex. Its primary clinical finding is lactic acidosis. Such PDCD mutations, leading to subsequent deficiencies in NAD and FAD production, hinder oxidative phosphorylation processes that are key in aerobic respiration. Thus, acetyl-CoA is instead reduced via anaerobic mechanisms into other molecules like lactate, leading to an excess of bodily lactate and associated neurological pathologies. While pyruvate dehydrogenase deficiency is rare, there are a variety of different genes when mutated or nonfunctional that can induce this deficiency. First, the E1 subunit of pyruvate dehydrogenase contains four different subunits: two alpha subunits designated as E1-alpha and two beta subunits designated as E1-beta. The PDHA1 gene found in the E1-alpha subunits, when mutated, causes 80% of the cases of pyruvate dehydrogenase deficiency because this mutation abridges the E1-alpha protein. Decreased functional E1 alpha prevents pyruvate dehydrogenase from sufficiently binding to pyruvate, thus reducing the activity of the overall complex. When the PDHB gene found in the E1 beta subunit of the complex is mutated, this also leads to pyruvate dehydrogenase deficiency.

However, Oliver Smithies made significant contributions. Bier states: "The method of Smithies ... is finding wide application because of its unique separatory power." Taken in context, Bier clearly implies that Smithies' method is an improvement.
=== Separating-funnel method === In this method the solid particles present into the two immiscible liquids can be easily separated by suspending those solid particles directly into these immiscible or somewhat miscible liquids.
==== 2012 Jalisco massacres ==== The dismembered remains of 18 bodies were found inside a Toyota Sienna and Ford EcoSport near the U.S. retiree communities in Chapala, Jalisco, just south of the city of Guadalajara. Eighteen heads were found along the dismembered bodies; some had been frozen, others were covered in lime, and the rest were found in an advanced state of decomposition. An anonymous call alerted the police to the abandoned vehicles, which were found by the side of a highway early in the morning on 9 May 2012. They were consequently towed to government offices to unload the bodies. The authorities confirmed that a message was left behind by the killers, presumably from Los Zetas and the Milenio Cartel. The attorney general of the state of Jalisco, Tomás Coronado Olmos, stated that this massacre was a revenge attack for the 23 killed in the 2012 Nuevo Laredo massacres. In addition, 25 people were rescued after being kidnapped in Tala, Jalisco on 8 May 2012; the killers had plans to kill and "throw" them for public display. Another 10 people managed to escape their capture by members of Los Zetas that same day, and alerted the local media of the situation. Upon the arrest of the four alleged killers, one of the cartel members confessed that they had plans to "repeat" what had happened in the 2011 Guadalajara massacre, where 26 bodies were dumped in a major avenue for public display. According to Proceso magazine, Los Zetas were planning to kill 50 people on 9 May 2012, a day before Mother's Day.

No. Melanotan-2 is manufactured synthetically. The naturally occurring peptide in the same family is alpha-melanocyte-stimulating hormone, which the body produces as part of normal endocrine and neural signalling.
Both are synthetic analogues derived from the same parent hormone. Reference descriptions usually characterise afamelanotide as more selective for the MC1R subtype, whereas melanotan-2 is reported to interact with a wider range of melanocortin receptors.
Early studies explored melanocortin signalling and pigmentation, which later led to informal use of the label tanning peptide. That label is not a regulatory category and does not imply approved status for any purpose.
Purity is normally stated as an area percentage from high-performance liquid chromatography, for example ninety-five or ninety-eight percent. That figure describes the proportion of ultraviolet-absorbing material eluting as the main peak. It says nothing about water content, counterions, residual solvents or mass fraction of the peptide itself.
